Nucleotide excision repair pathway assessment in DNA exposed to low-intensity red and infrared lasers BJMBR

Low-intensity lasers are used for prevention and management of oral mucositis induced by anticancer therapy, but the effectiveness of treatment depends on the genetic characteristics of affected cells. This study evaluated the survival and induction of filamentation of Escherichia coli cells deficient in the nucleotide excision repair pathway, and the action of T4endonuclease V on plasmid DNA exposed to low-intensity red and near-infrared laser light. Occurrence of adhesin-encoding operons in Escherichia coli isolated from breeders with salpingitis and chicks with omphalitis BJM

The occurrence of fim, pap and sfa operons in Escherichia coli isolated from breeders with salpingitis and chicks with omphalitis was evaluated. Analysis of 100 E. coli isolates, by colony hybridization tests, showed that 78 (78%) were fim+, one (1%) was sfa+, seven (7%) were fim+ associated with pap+, eigth (8%) were fim+ and sfa+, one (1%) was fim+pap+sfa+ and five (5%) isolates did not hybridize with any probe. These results suggest that fim adhesion-encoding operon plays an important role in pathogenesis of E. coli infection in chickens with salpingitis and omphalitis.

Occurrence of non-O157 Shiga toxin-producing Escherichia coli in dogs with diarrhea Ciência Rural

Shiga toxigenic Escherichia coli (STEC) and Attaching and effacing E. coli (AEEC) have been associated with diarrhea illness in dogs. From January to December 2006, 92 E. coli isolates from 25 diarrheic dogs were analyzed, by screening for the presence of Shiga toxin-producing (stx 1 and stx 2) and intimin (eae) genes. Twelve isolates were detected by PCR to harbor the Shiga toxin genes (7 the stx 1 (7.6%); 5 the stx 2 (5.4%); and none both of them). Occurrence of toxigenic Escherichia coli in raw milk cheese in Brazil Arq. Bras. Med. Vet. Zootec.

The occurrence of toxigenic Escherichia coli in raw milk cheese was surveyed in Middle Western Brazil. Fifty samples of cheese from different supermarkets were analyzed for E.coli. The isolates were serotyped and screened for the presence of verotoxigenic E. coli (VTEC) and enterotoxigenic E. coli (ETEC) by Polymerase Chain Reaction (PCR). The susceptibility to thirteen antimicrobial agents was evaluated by the disk diffusion method. E.coli were recovered from 48 (96.0%) of the samples. A análise de Escherichia coli, considerada bactéria indicadora de contaminação fecal, é utilizada na verificação do controle de processos de abate. Porém, poucos trabalhos foram publicados acerca da ocorrência desse micro-organismo nas indústrias de abate brasileiras. Assim, o presente estudo determinou a ocorrência de E. coli genérica em carcaças bovinas em um estabelecimento sob inspeção federal, habilitado à exportação, identificando as possíveis variações no ano de 2010. The cloning, expression and purification of the glutathione (sulfur) import system ATP-binding protein (gsiA) was carried out. The coding sequence of Escherichia coli gsiA, which encodes the ATP-binding protein of a glutathione importer, was amplified by PCR, and then inserted into a prokaryotic expression vector pWaldo-GFPe harboring green fluorescent protein (GFP) reporter gene. The resulting recombinant plasmid pWaldo-GFP-GsiA was transformed into various E. coli strains, and expression conditions were optimized. O objetivo desse trabalho consistiu em determinar a patogenicidade in vivo e in vitro de 25 amostras de Escherichia coli obtidas de aves com Síndrome da Cabeça Inchada (SCI). A patogenicidade in vivo foi determinada pela inoculação em pintos SPF de um dia de idade, utilizando-se quatro vias de inoculação: intracardíaca, saco aéreo, oral e traqueal. As aves foram avaliadas quanto à mortalidade a cada 12 horas, por um período de sete dias. Os resultados desse estudo demonstraram que as 25 amostras de E. coli, isoladas de aves com SCI, causaram mortalidade mais elevada em pintos de um dia de idade quando foram inoculadas pelas vias intracardíaca (73,6%) e saco aéreo (68%). Phylogenetic grouping and pathotypic comparison of urine and fecal Escherichia coli isolates from children with urinary tract infection BJM

The aim of this study was to investigate the phylogenetic background and to assess hlyD (involved in the secretion of haemolysin A) and intll (encoding a class 1 integrase) in Escherichia coli isolates derived from urinary and fecal specimens. A total of 200 E. coli isolates was collected from patients presenting with urinary tract infection (UTI) during September 2009 to September 2010 and screened for hlyD and intll genes by polymerase chain reaction (PCR). O objetivo deste relato foi descrever um caso de piometra em uma leoa (Panthera leo) de cativeiro, as lesões de necropsia e histológicas, bem como os resultados da análise microbiológica. Abstract Multidrug-resistant microorganisms are of great concern to public health. Genetic mobile elements, such as plasmids, are among the most relevant mechanisms by which bacteria achieve this resistance. We obtained an Escherichia coli strain CM6, isolated from cattle presenting severe diarrheic symptoms in the State of Querétaro, Mexico. It was found to contain a 70 kb plasmid (pMEX01) with a high similarity to the pHK01-like plasmids that were previously identified and described in Hong Kong. Pneumonia as the cause of death in gilts experimentally infected with Trypanosoma brucei OAK

A study was conducted in 24 gilts to determine the effect of Trypanosoma brucei infection on their reproductive efficiency. The infected gilts developed clinical trypanosomosis following a prepatent period of 2-3 days with 1.8 x 10⁶ trypanosomes per gilt. The clinical signs were observed intermittent fever, pale mucus membranes, short moist cough, moist rales, mucopurulent ocular discharges and hyperemia of the skin, reduced feed intake, and loss of body condition, recumbency, uncoordinated movements, posterior paresis and death of gilts. Susceptibility to several ethanolic extracts of propolis (EEP) concentrations was tested with the population analysis technique in reference strains of Staphylococcus aureus (ATCC 25923) and Escherichia coli (ATCC 35218). The results of these tests showed that all bacteria were killed by EEP concentrations approximately equal or higher (2.0% v/v- S. aureus; 10.0% v/v - E. coli) than the respective minimal inhibitory concentrations (MIC). Regarding the susceptibility to propolis, there was a homogeneity of data with the respective time kill curves showing a clear bactericidal effect during 6 to 9 h of exposition.

Practical use of immobilized lysozyme for the remediation process of Escherichia coli in aqueous solution Electron. J. Biotechnol.

The lysozyme enzyme was immobilized on vitreous surface (fragments with diameters of 0.3 and 1.0 mm) for remediation of the microorganism Escherichia coli JM 109 into fresh water and saline solutions with 0.9% NaCl (w/v). Characterization of enzymatic film was carried out by infrared spectroscopy and atomic force microscopy techniques. Bactericide activity of the enzyme was evaluated by spectrophotometric analysis. It was verified that the enzymatic film was strongly coupled with the vitreous surface. The topographic analysis demonstrated that the deposited film was uniform and homogeneous. Presence of extraintestinal pathogenic Escherichia coli in butcheries in Taquaritinga, SP, Brazil BJM

The study was conducted in twenty-three butcheries in the city of Taquaritinga, State of São Paulo, Brazil, surveyed during a 10 months period. Among two hundred and eighty-seven Escherichia coli strains isolated from samples of ground beef, meat-grinding-machines and the hands of manipulators, five were recognized as extraintestinal pathogenic E. coli (ExPEC), showing virulence factors (P and S fimbriae, hemolysin and aerobactin) and presenting multidrug resistance. Retail-sold food may constitute an important vehicle for the dissemination of ExPEC in communities, giving rise to reasons for concern.

El objetivo del presente trabajo fue determinar la frecuencia de algunos factores de resistencia antibiótica de Escherichia coli en plantas Tipo Inspección Federal (TIF) del Estado de México. This study aims to assess the prevalence of Shiga toxin-producing Escherichia coli (STEC) in ground beef collected in two cities located in the State of São Paulo, Brazil. A total of 250 samples of raw ground beef were collected in local grocery stores during the period of March to December 2002 in the cities of Ribeirão Preto (114 samples) and Campinas (136 samples), São Paulo State, Brazil. Prevalence of diarrheogenic Escherichia coli and rotavirus among children from Botucatu, São Paulo State, Brazil BJMBR

In a one-year prospective study carried out to define the role of rotavirus and Escherichia coli in local childhood diarrhea, we determined the prevalence of both agents in 54 diarrheic children attending a health center in Botucatu.
